After a Calm Mind, Serperior's Special Attack will only be as high as a neutral natured Jellicent's, so getting off more than one Calm Mind should be your goal, and is safer when Serperior has Rest and Chesto Berry. With a Chesto Berry equipped to it, Serperior wakes up if it is asleep then the Chesto Berry gets consumed.

With Overgrow, at 1/3 or less of its max hp Serperior's offensive stat is 1.5x with grass attacks. This Moveset is a tricky one. I can't decide for its Nature though. First is to set Leech seed + Growth combo. Once Special attack increased, use Giga Drain and your HP will be boosted to Full then use Wring Out another Special Attack to achieve its maximum damage dealt which is 120. This is a no TM and no Breeding moveset so any can replace Growth with Calm Mind though. With a Big Root equipped to it, Serperior gains 1.3x hp from draining moves, aqua ring, ingrain, and leech seed.
